You should go swimming at Folly Beach, and have an early dinner at The Anchor Line. You should walk on the battery at sunset, and then have dessert at Kaminski’s.

My family’s tradition is to eat at The Crab House once while there. I’ve written an entry about it here The Trawler and California Dreamin’ are also good restaurants.
If you like history, then Charleston is going to be magical to you. There are walking tours you can take about the historical downtown area. There are several historical homes in the area. I like Boone Hall Plantation. You cannot truly go to Charleston without going to the historical downtown area and seeing The Battery and Rainbow Row, in my opinion.
As mentioned in one of the above posts, Folly Beach is good if you like the beach/ocean.

My favorite thing to do is to walk around the city. You can go to the visitor center at 375 Meeting St. and pick up a walking map. From the visitor center take the trolly into downtown and start from there. If you can get tired, on Market St. by the open market you can take a carriage ride. It’s a bit pricey, but local stores have discount tickets. It’s a one time thing to do, but it’s nice and relaxing listening to the history of the city. My favorite beach to visit is Isle of Palms, more relaxing than Folly Beach in my opinion. A favorite place of ours to eat is Poogan’s Porch at 72 Queen St. close to the market area. It’s already been said in the other replies but I’ll say it again; walk around and explore the city. Oh, and as has also already been said, if you like the ocean, go to Folly Beach.
