Visit the Reichstag. It’s beautiful at sunset. And the Potsdamer Platz, too, especially at night. I love the architecture.
Charlottenburg Palace is worth a visit, if you’re into castles and nice gardens. There are a few smaller museums on site, including the Palace itself and a Mausoleum in the park.
At Hackesche Höfe you’ll find some very nice backyards, offering great eating places, smaller shops (or so I’ve heard), and a few things to do at night.

For me, one of the most powerful parts of my visit was the evening I spent in the Jewish Museum of Berlin. I wholeheartedly recommend spending a few hours there. It’s exceptionally well done and very informative.
Also second the recommendation for the Reichstag and would like to add Wilhelm-Kaiser church (pictured in my comment). It’s near the ‘Zoo’ station.

Make sure you check out the Checkpoint Charlie Museum. Its awesome! Plus the thing SaraEMiller posted a picture of is pretty cool too, but I still recommend the museum more. Its lesser known, but for me its a must-see.

Definitely the Reichstag, Haus am Checkpoint Charlie (Museum) and die Gedaechnis Kirche – so much history there. Also for shopping/people watching – KaDeWe near Wittenberg Platz station and the KuDamm – Berlin’s answer to NY 5th Avenue or Paris’ Chans elysees. Beautiful/easy hiking in Gruenewald or out in Potsdam at Sansouci Castle- about 30 minutes or so drive, if I remember right. Be sure to find a sidewalk cafe almost anywhere in the city and watch the world go by. Some of the best people watching around. Also – must have a Currywurst at some roadside Imbiss. Yummy. If your German’s descent, take a boat tour on the Spree. It’s a different look at some of the same places, but you get to go behind & between for a totally different look at a few centuries of the city’s architecture and views. Very cool. I think some of the tours are also in English/French/Spanish/what have you. Gute Reise!!

And since you are in Berlin I guess you could have a visit to Potsdam too. It’s definitely worth it. and also Spree forest at Lubbenau!
and a must do is a walk around Gendarmenmarkt, Unter den Linden and Potsdamer Platz, then have dinner in the city!

The Spreewald is in deed a very wonderful place to visit, but it’s quite a distance from Berlin and not really a Berlin experience. They have incredible pickles, however and on Father’s Day, the forest is jammed with men on floating rafts topped with picknick tables drinking beer and schapps, singing old songs, wearing flowers on their lappels. The Spreewald Gurken is mentioned in the movie, “Goodbye Lenin” as a specialty of East Germany.
If one is willing to travel to the Spreewald, then you should also get to Werder. Werder is famous for it’s fruit wines and they have a festival every May where you can easily sample hundreds.

besides the usual places others have mentioned,i greatly enjoyed the Judisches Museum and the whole walk down the very long Unter den Linden.
checkpoint charlie is quite a landmark. just note that it is usually overrun with tourist.
you’d enjoy Walking around berlin too, its a good way to see the raw berlin in between tourist spots; alexanderplatz has a good new buzz about it.

You’ve got to visit the Jewish Memorial (near the Reichstag), and its close to the Brandenburger Tor, all things you ought to see. I definitely agree that you should visit the Jewish Museum too. And the Fernsehturm gives you a great view over all of Berlin.

For me, I would say that the must-go place would be the museam scene on Museaum Island in the middle of the Spree. The museums are all excellent, and cover a wide variety of subject-matter. The best part… they are open late (until 10 pm) on Thursday evenings, and are completely free from 6 pm until 10 pm on Thursdays.

I didn’t get a chance to do this but go up the Fernsehnturm (?) – it’s a cool looking spire which can be seen all over Berlin – go up and have a drink and fab views! (apparently!)

Wow, Berlin has such a vast amount of things to do and see, it is an extremely difficult thing to name simply one.
In spite of that, I would say that a guided walking tour of the city is the must-do-thing. My first trip to Berlin was in 2000. I took a walking tour that started at Zoologischer Garten S-Bahnhof. It gave us a great overview of the city’s landmarks and history. From the starting point, we S-Bahn’d to Hackescher Markt, then meandered our way over to Friedrichstr. and through the Museum Island area. We were all over and the feet hurt afterwards, but our orientation was enhanced 100-fold. After that, you can certainly know where you would like to further explore.
The must-go-place, IMHO, would be Kollwitzplatz on Sunday morning for a Brunch. It’s bustling with people, in the christmas time there is also a Weinachtsmarkt. The restaurants provide all you can eat brunches, some more expensive than others, but all very delicious. Brunch on Sunday morning is a quintessential Berlin experience.
If you are a clubber, than the Icon club is a must-go-place. Saturday nights are drum and bass night and the house DJs are some of the best in Berlin.

I spent several months in Berlin, and it is really a wonderful city. Still, my favorite place (and the one I went to most frequently) was the Reichstag Building. The tour is really interesting, but if that isn’t your thing, head strait for the dome—it is the most beautiful view of Berlin in the city. Best of all, it is free, so you can save your Euros for beer and pretzels.
Oh, and eat lots and lots of apple cake (apfel kuchen)!

Try also Kreuzberg. Nice place to have a good evening. Visit one of the real small bars, where only five or six people can sit at the counter.
